aim:localizeddeliveryofgrowthfactorshassignificantpotentialasafuturetherapeuticstrategyintissueengineeringandregenerativemedicine.ananoparticlevehiclewascreatedandevaluatedinthisstudywiththeintenttodelivergrowthfactorsforperiodontalregeneration.
methods:novelcompositenanoparticlesbasedonglycidylmethacrylatederivatizeddextrans(dex-gma)andgelatinwerefabricatedbyafacilemethodwithoutusinganyorganicsolvents.theconfigurationsoftheresultantnanoparticleswereevaluatedbytransmissionelectronmicroscopy,scanningelectronmicroscopy,andatomicforcemicroscope.theirsurfaceswerecharacterizedbyzeta-potentialmeasurements,afterwhichtheirpropertiesincludingswelling,degradation,drugrelease,andcytotoxicitywerealsoinvestigatedusinginvitromodels.
results:theparticlesizeofdex-gma/gelatinnanoparticles(dg-nps)rangedfrom20to100nmandshowedamono-dispersesizedistribution(meandiameter53.7nm)andastronglynegativesurfacezetapotential(−20mv).thedg-npswerecharacterizedbygoodswellinganddegradationpropertiesinmediaincludingdextranase.theinvitrodrugreleasestudiesshowedthattheefficientbonemorphogeneticprotein(bmp)releasefromdg-npswasmaintainedformorethan12dunderdegradationconditions,wheremorethan90%oftheloadedbmpwasreleased.noanyrelevantcelldamagecausedbydg-npswasfoundinthecytotoxicitytestsforaperiodof24h.
conclusion:thesecombinedresultsdemonstratethatdg-npsfulfillthebasicprerequisitesforgrowthfactordelivery.withfurtherinvivostudies,thosenanoparticlesmayofferapromisingvehicleforthedeliveryofactivedrugstotheperiodontium.
